What Is MOTS-C?

MOTS-C (mitochondrial open reading frame of the 12S rRNA-c) is a 16-amino acid peptide encoded within the mitochondrial genome — specifically the 12S ribosomal RNA gene. Discovered in 2015 by Lee et al. at the University of Southern California, it represents a new class of mitochondria-derived peptides (MDPs) with systemic signalling functions.

Unlike most peptides encoded in nuclear DNA, MOTS-C originates from the mitochondrial genome, placing it at the intersection of mitochondrial biology and systemic metabolic regulation. Circulating levels of MOTS-C decline with age, drawing interest from longevity researchers.
